PHOTOGRAPHY WITH NIKON MICROSCOPE IN ANIMAL FACILITY
Film: ASA 50-400
- Black and white: T-Max 400
- Color slides: Ektachrome 400 or Fujichrome 400
- Stained sections: Fijichrome 50 works well
Procedure
- Set brightness control dial to:
| daylight film |
9 |
| tungsten light |
8 |
| black and white |
6 or higher |
- Spot meter reads 1% of area within double cross line
- Average (large circle) averages 30% of area
- Exposure adjust: 1/3 EV steps from -2 to -~
- +1 EV + extends shutter time
- - decreases shutter time
- -1 = about 1/2 exposure time
- -2 = about 1/4 exposure time
- For fluorescence microscopy with extremely dark background:
- 0 exposure adjustment if bright area of specimen covers area for spot
meter
- - adjust if does not cover spot
- It is best to make slides and have prints made directly from slides if desired.
